Prologue plays during the game's introductory story cut scene where a zeppelin descends on New York home. It continues as the messenger meets Nemo at his bedroom window to invite him to Slumberland to play with Princess Camille, and their ride to Slumberland.

In an interview, Junko Tamiya had this to say about Prologue:

Nemo featured a genre I had not dabbled in before, but I made something fresh and was able to get everything done. For example, for the opening theme, I conveyed the sound of an amusement park with a barrel organ and incorporated distant waves and fermatas, among other musical features.
